If I make a questionnaire available to a colleague via hyperlink and then make changes to that questionnaire, do I need to update the hyperlink?

That is not necessary. Any changes to a questionnaire (e.g.: removing clauses, adding questions, changing card order, etc…) automatically show up in the hyperlink that was created before the changes were made.
